▼a Abl family kinases are believed to play as central signaling nodes to coordinate microtubule dynamics by transducing signals from upstream guidance receptors to downstream microtubule-associated proteins. Here, I report a novel microtubule dynamics regulation mechanism via direct binding of Abl2. In this study, I discovered that Abl2 directly binds microtubules and regulate microtubule dynamics. In a purified recombinant system, I identified a central proline-rich region inside Abl2 that is sufficient and necessary to bind to microtubules, and the interaction between Abl2 and microtubules is electrostatically mediated by the COOH-terminal acidic tubulin tail. Using in vitro single-filament microscopy, I found that Abl2 stably bound onto microtubule lattice and regulated microtubule dynamics by increasing elongation rate, reducing shortening rate, and decreasing catastrophe frequency. In cells, when Abl2 was knocked out microtubule growth rate was significant reduced and re-expressing Abl2 successfully rescued the microtubule growth defect. These and other data suggest a model where Abl2 act as a microtubule regulator at the cell periphery. |
