요약 | Once considered mere noise, fMRI has become a major neuroscience tool in part due to early studies demonstrating its utility in mapping fundamental functions in the human brain. This is a relatively young branch of science, and exciting new techniques often emerge and disseminate before we can fully understand their limitations. Recent work has raised questions about the reliability and validity of major fMRI statistical methods that are both habitually underpowered and nonspecific. This work seeks to address these unresolved questions. The first three content chapters focused on influences on the reliability of functional connectivity. The first chapter reveals that functional connectivity acquired across multiple sites across North America did not show major site, scanner manufacturer, or day effects. In the second chapter, a unique, extensively sampled dataset was used to address how amount of data per subject influences whole-brain reliability, the influence of increasing runs versus sessions, the spatial distribution of reliability, the reliability of multivariate methods, and, crucially, how reliability maps onto utility. The third chapter contains the first ever meta-analysis and systematic review of the literature, which underscored the poor reliability of functional connectivity, the conditions under which reliability improves (both spatial and processing), and limitations of the measure.
