요약Obesity is a prevailing issue across the globe. Women who are obese often suffer from delayed onset of lactogenesis and tend to breastfeed for shorter durations. Breastfeeding is extremely important in infant development and has been demonstrated to decrease incidences type 2 diabetes and asthma later in life. Woefully, lactation is an understudied field and very little is known about the complexities of genotype, diet, and their subsequent lactation outcomes. Peripheral serotonin is becoming more well known as an anti-obesity target and is a large focus of this thesis. Additionally, we utilize the low-density lipoprotein receptor knockout (Ldlr-KO) mouse, as an exacerbated pro-obesity and hypercholesterolemic model. We also generated a Tph1/Ldlr KO (double knockout/DKO) model with the hopes of understanding how a pro-obesity and anti-obesity gene may interact. Chapter 2 demonstrates that during peak lactation, dams on a high-fat diet (HFD) have alterations in collagen biosynthesis, inflammation, and immune pathways. Additionally, consumption of a HFD altered mRNA splicing and isoform expression in the lactating mammary gland. Chapter 3 gives a cursory overview to how Tph1 responds to a LFD and HFD when in comparison to wildtype (WT) mice during peak lactation. Illustrated in Chapter 4 are the complexities between genotype and diet in relation to lactation. We characterize descriptive changes during early and mid-lactation, but it is not mechanistic in nature. Chapter 5 utilizes RNA-sequencing in order to get a better global overview of how varying genotypes have exclusive and interactive functions during lactation and demonstrates potential pathways of interest for us to explore in the future. Finally, chapter 6 utilizes a peripheral serotonin inhibitor during the peripartum period to emphasize the importance of conducting pharmacologic agents at all points in development and how inhibition does not necessarily mirror a transgenic mouse model.
