요약Bacteria use sigma factors to regulate and control gene expression. Extra cytoplasmic function sigma factors are activated during times of stress to enhance the survival of the cell. Here we show that pBS32, a large, low copy number plasmid present in the ancestral strain of Bacillus subtilis, encodes an extra cytoplasmic function sigma factor, SigN, that mediates cell death after DNA damage. Using molecular biology, biochemistry, and genetics, SigN was characterized. We show that SigN interacts with RNAP to initiate transcription, is repressed by LexA, autoactivates, and contains a regulon on pBS32. The mechanism of death remains unknown, but we know that the global transcriptional repressor, AbrB, is needed for death to occur. With these new findings, we have increased the knowledge base of pBS32 and expanded what we currently know about extra cytoplasmic function sigma factors.
