요약Vagal tone-a cardiac measure of parasympathetic nervous system function-correlates with improved stress response, illness recovery, and numerous other physiological functions. Most of the research investigating vagal tone in mice has been performed with adult subjects in room-temperature environments. Such studies have not proliferated with the increased popularity of mice in biomedical research because it is generally thought that mice do not exhibit vagal tone (Janssen & Smits, 2002). Recent studies, however, have demonstrated that adult mice will exhibit vagal tone at warm ambient temperatures. Nevertheless, little is known about how vagal tone develops. My work was designed to assess the development of vagal tone in mice through a series of five studies. In the first study, heart rates of mouse pups in three age ranges (Postnatal Days [PD] 6-8, 10-12, and 16-19) were recorded under cool (23 째C), warm (32 째C), and thermoneutral (36 째C) ambient temperatures. As with adult mice, the oldest group exhibited vagal tone at the warmest temperatures, however, none was observed in any other group. Under the same recording conditions as the first study, the second and third studies investigated the development of the parasympathetic nervous system by pharmacological block of the sympathetic nervous system and vice versa. The fourth study investigated the potential modulation of vagal tone via administration of an anxiolytic stimulus (a familial odor). No effect on vagal tone was observed despite the known relationship between stress responses and vagal tone in humans. The fifth study successfully validated my methods by replicating the results of published adult data. Combined, these data represent the first evidence of vagal tone in the preweanling mouse. Furthermore, these findings suggest that housing mice at thermoneutral temperatures may produce more human-like physiological states, which may then serve as better models for translational research.
