요약High-density arrays have been a crucial tool for the biosciences for more than 25 years. DNA array technology has been applied to nucleic acid detection, the characterization of binding interactions, and gene synthesis. Two strategies for synthesizing arrays on flexible substrates are described here, as well as the capabilities of a maskless DNA array synthesizer for producing arrays of RNA. Array synthesis chemistry was developed so that arrays of DNA could be made upon flexible sheets of poly(ethylene terephthalate) (PET) coated with crosslinked polymer bilayers. Arrays made upon the bilayer-coated substrates are suited for array applications that require relatively short oligonucleotides. The use of uncoated PET was examined and was found to be suitable for fabricating arrays comprised of longer oligonucleotides. Both approaches enable the arrays to be divided into subsections after synthesis in order to increase array fabrication throughput. A strategy for fabricating arrays of RNA was explored, which enables new enzymatic assays and reveals avenues for improving array quality. The work described here addresses the major fields of array usage and suggests many promising routes to further develop the technology.
